잿빛곰팡이병 약제 저항성균주 판별 kit 제작 및 활용방법


1. 활용

가. 매뉴얼에 따라 잿빛곰팡이병 약제 저항성균주 판별 kit 제작 

나. 제작된 kit에 꽃잎을 치상하거나 낙하포자 채집 

다. 20℃항온기에서 배양 후 저항성균주 판별하여 적용약제 선택 방제


2. 현황 및 문제점

가. 잿빛곰팡이병에서 약제저항성균주의 출현으로 기존약제의 방제가가떨어지고 있음

나. 토마토 재배농가들이 자신의 포장에 적합한 약제를 처방 할 수 있는 방법이없어 관행적으로 농약을 사용함으로써 경영비 부담 및 농약오염 문제 발생

4. 기대효과

가. 약제에 내성이 생긴 잿빛곰팡이를 효과적으로 방제함으로써 농가 소득 증대

나. 살균제를 적게 사용함으로써 안전한 토마토 생산


5. 개발기술의 활용방법

<잿빛곰팡이 약제 내성균주 판별 kit 제작 방법>

가. 배지종류 : 잿빛곰팡이 선택배지(Botrytis Selective Medium(BSM))

잿빛곰팡이 포자 선택배지(Botrytis Spore Trap Medium(BSTM))

나. 약제내성균주 kit 제작

-  잿빛곰팡이 선택배지(BSM)

glucose : 2g,   NaNO3 : 0.1g,  K2HPO2 : 0.1g

MgSO47H2O : 0.2g, chlorampenicol : 0.2g

pentachloronotrobenzene : 0.02g, Maneb 80 : 0.02g

rose bengal : 0.05g, tannic acid : 5g, agar : 20g/L

※ autoclaving 하지 말고 끓여서 배지 만들 것, pH는 4.5로 교정(NaOH 이용)


-  잿빛곰팡이 포자 선택배지(BSTM)

glucose : 2g,   NaNO3 : 0.1g,  K2HPO2 : 0.1g

MgSO47H2O : 0.2g, chlorampenicol : 0.2g

pentachloronotrobenzene : 0.02g, Maneb 80 : 0.02g

Rubigan : 0.1㎖, tannic acid : 5g, agar : 20g/L

※ autoclaving 하지 말고 끓여서 배지 만들 것

-  BSM or BSTM + 시판약제

‧ 약제 : Diethofencarb+Carbendazim, Benomyl, Fluazinam, Triflumizole Vinclozoline, Diethofencarb, Boscolid 등

‧ 선택배지에 농약을 혼합하여 제작(농약 혼합농도는 농약사용지침서 참조)

※ 선택배지가 식은 후 추가적으로 약제 혼합


<잿빛곰팡이 약제 내성균주 판별 kit 사용 방법>

가. 잿빛곰팡이 선택배지를 이용한 kit 사용

-  착과 후 죽은 꽃을 잿빛곰팡이 선택배지 위에 올려 놓는다

※주의사항 : 새로운 꽃을 배지위에 올려 놓을 때 화염소독 실시

-  28℃항균기에서 5~7일 정도 배양

-  약제가 혼합된 선택배지에서 잿빛곰팡이가 배양되지 않은 약제를 사용하여 방제

나. 잿빛곰팡이 포자 선택배지를 이용한 kit 사용

-  약제 가 혼합된 선택배지를 토마토 식물체 밑에 놓는다.

-  28℃항균기에서 5~7일 정도 배양

-  약제가 혼합된 선택배지에서 잿빛곰팡이가 배양되지 않은 약제를 사용하여 방제
